Isolation and diagnosis of Sinorhizobium meliloti symbiont of alfalfa with the study of effect salinity and pH on it

Sixty local isolates of Sinorhizobium melilote.were isolated from Medicago sativa root nodules.The colonies characteristics were studied after growing on trypton yeast extract agar (Ty).All isolates were gram negative, rod shape and motile because of formation of cyclic B-(1-2)glucans.All isolates showed ability for production of cellulose fibrils and nonproduction of cyclic B-(1-3)glucans.The bacterial isolates formed nodules on infected plants after two weeks .Adding of differ at antibiotics to the cultural media in different concentration lead to inhibit the growth of some of the bacteria isolates.All isolates showed agood growth on media having 2% and 3% NaCl, but the growth was inhibited at 7% NaCl.regarding K2SO4 all isolates shawed growth on media containing 2%,3% and 4%K2SO4 but no growth was observed on 7.5%K2SO4 .pH 4 inhibit the growth of all isolates in contrast there was agood growth at pH ranging for 6 to 10.The total nitrogen of plant inoculated with bacterial isolates were determined of the soil characteristics were recorded.

The present study is included preparation of aqueous & acetonic plant extracts for Tagete patula , Ammi visnaga L and Convolvulus arvensis L. and tested their biological activity against Staphylococcus aureus , Streptococcus pyogenes , Escherichia coli and Klebsiella pneumonia by using five concentrations( 500 mg/ml , 250 mg/ ml ,125 mg/ml,0.06 mg/ml and 0.03 mg/ ml).
The aqueous extract of Convolvulus arvensis L. was not appeared any biological activity against tested isolates while the aqueous extracts of Tagete patula and Ammi visnaga L showed inhibition activity.
The acetonic extracts of the three plants showed inhibition zone but the highest inhibition diameter rate was (28 mm) for Ammi visnaga L at 500 mg/ml .
The chemical tests for all plant extracts performed for identified active compounds.

Salt tolerance of two rice( Oryza sativa L.) Cv. cultured in vitro

Series of experiments were conducted at Tissue Culture Laboratory. at Al- zubair ,Basrah to
study effects of salinity on rice Oryza sativa L. cultivars named Anbar and Jasmine , cultured in vitro .
Mature seeds (mature embryo + endosperm ) were used as explants which cultured with MS salts basal medium supplemented with sucrose , vitamins , amino acids and the auxin 2,4-D.
Sodium chloride (NaCl) was added at concentrations (0 , 1 , 2 , 3 )%.
Results showed that increasing NaCl concentration decreased in callus fresh weight significantely
specially at 2% and 3 %.
Results also indicated that increasing NaCl concentration caused a significant increase in Na+and
proline concentrations , while it caused a significant decrease in K + and KNa ratio in the callus and
were a signifcant differences between the two varaieties in their salt tolerance

Study of leaves spot on Date Palm plantlets Phoenix dactylifera L.

This study was conducted of Date Palm Research Centar ,Basrah University year
(2009 – 2010 ) , to
determined leaf spot cause observed on leaves plantlets of date palm (tissue culture ) of
Barhee cultivar .
The fungus Alternaria alternata was isolated from leaves plantlets with black spot symptoms , Pathogenicity test showed to ability fungus A.alternata to gave spot sympyoms on treat leaves plantlets .
Showed the fungus Trichoderma harzianum ability in the inhibition of fungal growth A.alternata.

Control the larval and pupal stages of Meat Fly Sarcophaga haemorrhoidalis (Fallen) (Sarcophagidae: Diptera) by using the phenolic and alkaloids extracts of Citrullus colocynthis

This study done to evaluate the efficiency of alkaloids and Phenolic extracts of Citrullus colocynthis to control the larvae and Pupae stages of meat fly Sarcophaga haemorrhoidalis .
Results revealed the surpass of phenols of C. colocynthis on the extracts of alkaloids insect and by the average reached to 40.7% opposite to 31.06% of the phenolic, The phenolic extracts give mortality rates reached 90% , 70% and 66.6% for the 1st , 2nd and 3rd instars on the concentration 50% , while they for the occurrence high percentage of mortality for larval stage of this reached to 70% , 56.6% and 50% for the larval three instars on alkaloids extracts on the same concentration . The first larval stage much effected by the extracts with mortality medium reached to 48.8% by treating the phenolic and 40% for alkaloids extracts, there after the second and the third stage respectively.
Also results pupa with light weight relatively, The phenolic extracts was apparently in surpass concerned the great effect of the usage of extracts on pupal stages of this inset dead pupa and some morphological deformed pupa, also product over alkaloids in this effect and with average weight reached to 1.43 gm for 20 pupa where as alkaloids extract with 1.68 gm.

Effect of Irrigation Water Quality of Al-Sidoor and Al-Thanaaeb Locations on Some Productivity Properties for Date Palm

This study carried out in Abu Al-Kaseeb area southern to Basra to study the effect of irrigation water quality of Al-Sidoor and Al-Thanaieb location on some productivity properties of Date Palm Phoenix dactylifera L. C.V. Berhi and Hillawi during the season 2009.
The result showed that the water quality of Al-Sidoor and Al-Thanaieb are caused hard salinity problems when it use for irrigation as classification of F.A.O., Al-Thanaieb location had sodium, Potassium Chloraide, Bicarbonates, Sulfat ion, Electrical conductivity(E.C.) and Sodium adsorption ratio(SAR). More than Al-Sidoor while Al-Sidoor location had significant increase in yield dry weight and leaf length, but the water content and Prolin content shower opposite results

Milk pasteurization by microwave and study its chemical and microbiological characteristics during different storage times

A study was included whole cow milk pasteurization by H.T.S.T high temperature short time using 72ºC temperature for 15 second by tradition method and compare with microwave pasteurization apparatus and proceeded the following tests:
- The chemical tests:The percentage of moisture ,fat,lactose,ash,protein,acidity,pH,TBA value,FFA value.
- Microbiological tests: The aerobic total count,psychrotrophic bacteria,coli form bacteria,Salmonella,Staphylococcus aureus.
The following results were obtained:
1 – The pasterurization by using microwave was strength of the oxidation dye to reduction in TBA value and fatty acids compared with traditional pasteurization and control sample>
2 –The microbiological characterstics after pasteurization was good ,microwave was significantly better than traditional pasteurization and control sample because it reduced the total microbial number and psychrotrophic.
3 – coliform bacteria and salmonella do not found in the milk which pasteurized by microwave and traditional method during different storge times.
4 – The milk pasteurization with microwave did not have phosphatase compared with traditional method that have 15 IU⁄L where as the unpasteurized milk have 30 IU⁄L.
